Отримання стійких до гербіциду фосфінотрицину трансгенних рослин пшениці сорту Зимоярка трансформацією in vitro

Двома методами генетичної трансформації отримано стійкі до фосфінотрицину рослини м’якої пшениці сорту вітчизняної селекції Зимоярка, які несуть ген bar бактерії Streptomyces hygroscopicus. Біолістичну трансформацію проводили вектором рАНС25, Agrobacterium-опосередкованурСВ203 у штамі GV3101. Обидва вектори крім селективного гена фосфінотрицинацетилтрансферази (bar) містили репортерний ген b-глюкуронідази (uidA) Escherichia coli. Первинними експлантатами слугували незрілі зародки. Методом полімеразної ланцюгової реакції (ПЛР) доведено перенесення трансгена bar у геноми регенерантів і відсутність зараження агробактерією. Експресію гена uidA підтверджено гістохімічним аналізом. Ефективність трансформації біолістичним методом становила 0,5 %, Agrobacterium-опосередкованим — 1,25 %, що дало змогу відібрати відповідно 3 і 12 трансгенних ліній. Це перше повідомлення про успішне отримання в Україні трансгенної пшениці, стійкої до гербіциду фосфінотрицину, із застосуванням культури in vitro.
